Abstract: “Plasmon” is a synonym for collective oscillations of the conduction electrons in a metal nanoparticle (excited by an incoming light wave), which cause strong optical responses like efficient light scattering. The scattering cross-section with respect to the light wavelength depends not only on material, size and shape of the nanoparticle, but also on the refractive index of the embedding medium. For this reason, plasmonic nanoparticles are interesting candidates for sensing applications. Here, two novel setups for rapid spectral investigations of single nanoparticles and different sensing experiments are presented.\r\n\r\nPrecisely, the novel setups are based on an optical microscope operated in darkfield modus. For the fast single particle spectroscopy (fastSPS) setup, the entrance pinhole of a coupled spectrometer is replaced by a liquid crystal device (LCD) acting as spatially addressable electronic shutter. This improvement allows the automatic and continuous investigation of several particles in parallel for the first time. The second novel setup (RotPOL) uses\r\na rotating wedge-shaped polarizer and encodes the full polarization information of each particle within one image, which reveals the symmetry of the particles and their plasmon modes. Both setups are used to observe nanoparticle growth in situ on a single-particle level to extract quantitative data on nanoparticle growth.\r\n\r\nUsing the fastSPS setup, I investigate the membrane coating of gold nanorods in aqueous solution and show unequivocally the subsequent detection of protein binding to the membrane. This binding process leads to a spectral shift of the particles resonance due to the higher refractive index of the protein compared to water. Hence, the nanosized addressable sensor platform allows for local analysis of protein interactions with biological membranes as a function of the lateral composition of phase separated membranes.\r\n\r\nThe sensitivity on changes in the environmental refractive index depends on the particles’ aspect ratio. On the basis of simulations and experiments, I could present the existence of an optimal aspect ratio range between 3 and 4 for gold nanorods for sensing applications. A further sensitivity increase can only be reached by chemical modifications of the gold nanorods. This can be achieved by synthesizing an additional porous gold cage around the nanorods, resulting in a plasmon sensitivity raise of up to 50 % for those “nanorattles” compared to gold nanorods with the same resonance wavelength. Another possibility is\r\nto coat the gold nanorods with a thin silver shell. This reduces the single particle’s resonance spectral linewidth about 30 %, which enlarges the resolution of the observable shift. \r\n\r\nThis silver coating evokes the interesting effect of reducing the ensemble plasmon linewidth by changing the relation connecting particle shape and plasmon resonance wavelength. This change, I term plasmonic focusing, leads to less variation of resonance wavelengths for the same particle size distribution, which I show experimentally and theoretically.\r\n\r\nIn a system of two coupled nanoparticles, the plasmon modes of the transversal and longitudinal axis depend on the refractive index of the environmental solution, but only the latter one is influenced by the interparticle distance. I show that monitoring both modes provides a self-calibrating system, where interparticle distance variations and changes of the environmental refractive index can be determined with high precision.
„Plasmon“ ist ein Synonym für eine kollektive Schwingung (angeregt von einer eintreffenden Lichtwelle) von Leitungsband-Elektronen in einem metallischen Nanopartikel, welche eine starke optische Wechselwirkung wie z.B. Lichtstreuung hervorruft. Der Streuquerschnitt in Abhängigkeit von der Lichtwellenlänge hängt dabei nicht nur vom Material, Größe und Form des Nanopartikels ab, sondern auch vom Brechungsindex des umgebenden Mediums. Aus diesem Grunde sind plasmonische Nanopartikel interessante Kandidaten für sensorische Anwendungen. In dieser Arbeit stelle ich zwei neue Aufbauten zur schnellen spektralen Untersuchung von einzelnen Nanopartikeln und verschiedene Sensorik Experimente vor. \r\n\r\nIm genauen basieren die neuen Aufbauten auf einem optischen Mikroskop, welches im Dunkelfeld Modus betrieben wird. Beim „fast single particle spectroscopy (fastSPS)“ Aufbau wurde die Eingangslochblende eines angekoppelten Spektrometers durch ein „liquid crystal device (LCD)“ ersetzt, welches als einzeln ansteuerbare elektronische Blende fungierte. Diese Verbesserung erlaubt erstmals die automatische und kontinuierliche Untersuchung von mehreren Partikeln zur selben Zeit. Der zweite neue Aufbau (RotPOL) nutzt einen rotierenden keilförmigen Polarisator und bildet die volle Polarisationsinformation von jedem einzelnen Nanopartikel innerhalb eines Bildes ab, welches die Symmetrie der Partikel und deren Plasmon Moden aufzeigt. Beide Aufbauten werden genutzt um Nanopartikelwachstum in situ auf Einzelpartikel-Ebene zu beobachten um quantitative Daten von Nanopartikelwachstum zu extrahieren. \r\n\r\nMithilfe des fastSPS Aufbaus habe ich das Überziehen von Gold Nanostäbchen in wässriger Lösung mit einer Membran untersucht und zeigte eindeutig die anschließende Erkennung von Proteinbindungen an diese Membran. Dieser Bindungsprozess führt zu einer spektralen Verschiebung der Partikelresonanz aufgrund des höheren Brechungsindex von Proteinen im Vergleich zu Wasser. Daher ermöglicht diese miniaturisierte adressierbare Sensorplattform die lokale Analyse von Proteininteraktionen mit biologischen Membranen als Funktion der lateralen Zusammenstellung von phasenseparierten Membranen. \r\n\r\nDie Sensitivität bei Änderungen des Brechungsindex des umgebenden Mediums hängt vom Aspektverhältnis des Partikels ab. Auf der Basis von Simulationen und Messungen kann ich die Existenz eines optimalen Aspektverhältnis-Bereiches (im genauen zwischen 3 und 4) für sensorische Anwendung präsentieren. Eine weitere Erhöhung der Sensitivität kann man nur mittels chemischen Modifikationen der Gold Nanostäbchen erreichen. Dies kann durch die Synthese eines zusätzlichen porösen Goldkäfiges um die Nanostäbchen ermöglicht werden, was in einer Erhöhung der Plasmon Sensitivität dieser „Nanorasseln“ um 50% im Vergleich zu Gold Nanostäbchen bei derselben Resonanzwellenlänge resultiert. Eine andere Möglichkeit ist die Gold Nanostäbchen mit einer dünnen Silberschicht zu ummanteln. Dies reduziert die Linienbreite des Einzelpartikel-Spektrums um rund 30%, was die Auflösung des beobachtbaren Wellenlängenverschiebung erhöht. \r\n\r\nDiese Silberbeschichtung ruft den interessanten Effekt hervor, dass die Ensemble Linienbreite reduziert wird, indem die Relation, die die Partikelform mit der Plasmonresonanz-Wellenlänge verknüpft, verändert wird. Diese Änderung, die ich „Plasmonic Focusing“ genannt habe führt zu kleineren Abweichungen der Resonanzwellenlängen für dieselbe Partikel-Größen-Verteilung, was ich experimentell und theoretisch zeige. \r\n\r\nIn einem System von zwei gekoppelten Nanopartikeln, hängen die Plasmon Moden der transversalen und longitudinalen Achse vom Brechungsindex des umgebenden Mediums ab, allerdings wird nur die letztgenannte Mode vom interpartikel Abstand beeinflusst. Ich zeige dass die Überwachung beider Moden ein selbst-kalibrierendes System liefert, bei dem man interpartikel Abstand Variationen und Änderungen des umgebenden Brechungsindex mit hoher Präzision bestimmen kann.
