Abstract: | In der konvergenten Totalsynthese von C/D-cis-Östradiol-Derivaten dient die Aza-Claisen Umlagerung als Schlüsselreaktion. Zur Optimierung der Stereoselektivität der Auxiliar-gesteuerten Reaktion wurde eine Reihe neuer Pyrrolidin-Auxiliare synthetisiert. Kurze Synthesesequenzen ermöglichen die Herstellung der Ausgangsmaterialien für die Umlagerung: Das γ,δ-ungesättigte Säurefluorid trägt in ε-Position den A-Ring des Steroids und das Allylamin setzt sich aus dem Pyrrolidin-Auxiliar und dem Allylrest zusammen, der zum Aufbau der C/D-Ringe des Steroids dienen soll. Die Kupplung beider Fragmente gelang in hoher Ausbeute (67-99 %) und hoher Stereoselektivität, je nach Wahl des Auxiliar, zu dem einen (d.r. 87/12) oder zu dem anderen (d.r. 8/91) anti-Umlagerungsprodukt. Die Aufklärung ihrer Stereozentren gelang über Röntgenkristallstrukturen von Umlagerungsderivaten. Nach Abspaltung des Auxiliars über eine Iodlactonisierung konnte durch den analytischen Vergleich der enantiomeren Iodlactone die Struktur aller anti-Umlagerungsprodukte aufgeklärt werden. Die Steroidsynthese wurde fortgeführt, indem nach aromatischer Bromierung der Iodlactone diese reduktiv mit Zink geöffnet wurden und die entstehende Carbonsäure zum Weinrebamid transformiert wurde. Der B-Ring des Steroids wurde über einen Halogen-Metall-Austausch intramolekular geschlossen. Die Tetraloncarbonylgruppe konnte allyliert werden und der C-Ring über eine Metathese Reaktion geschlossen werden. Nach Abspaltung der Alkoholschutzgruppe und Oxidation zum Aldehyd, konnte der D-Ring des Δ9(11)-/D-cis-Methylöstradiol mittels radikalischer Cyclisierung in der gewünschten Regio- und Stereoselektivität fertig gestellt werden. During a convergent total synthesis of C/D-cis-estradiol derivatives the Aza-Claisen rearrangement serves as key reaction. To optimise the stereoselectivity of the auxiliary-directed reaction several new pyrrolidine auxiliaries were synthesized. Initial short sequences enabled to generate the starting materials for the rearrangement: The γ,δ-unsaturated acid fluoride bearing the steroids A-ring in ε-position and the allylamines composed of the pyrrolidine auxiliary and the allylic chain that will make up the steroids C- and D-rings. Coupling of the two fragments was accomplished during rearrangement in high yield (67-99 %) and with high stereoselectivity to either of the two antidiastereomers (d.r. 87/12 or 8/91) depending on the choice of auxiliary. The stereoconfiguration of the rearrangement products were elucidated by x-ray analysis of its derivatives. After cleavage of the auxiliary by iodolactonisation the structures of all anti-products were deduced by analytical comparison of the enantiomeric iodolactones. The synthesis of estradiol was continued by aromatic bromination and zinc-mediated ring-opening of the iodolacton. The so generated acid was transformed to its Weinreb amide that enabled the steroids B-ring closure by halogen-metal exchange. Allylation of the tetralone enabled the C-ring closure by metathesis reaction. After deprotection and oxidation of the alcohol the subsequent radical D-ring cyclisation completed the totalsynthesis of Δ9(11)-C/D-cis-methylestradiol in the desired regio- and stereoselectivity. |
