Abstract: | Bei den kupferhaltigen Hämocyaninen der Arthropoden handelt es sich um große,
extrazelluläre respiratorische Proteine, die sich aus mehreren Untereinheitentypen
zusammensetzen, und Hexamere (1x6-mere) bzw. Oligohexamere (2x6-mer,
4x6-mer, 6x6-mer, 8x6-mer) bilden. Für detaillierte Aussagen über die Funktionsweise
bei der reversiblen O2-Bindung dieser Proteine benötigt man genaue Kenntnisse über
deren dreidimensionale Struktur. Für die Aufklärung der Quartärstruktur eines solch
großen Proteins ist die 3D-Elektronenmikroskopie hervorragend geeignet, da sie
detaillierte 3D-Rekonstruktionen liefert. Damit wurden in dieser Arbeit die
Quartärstrukturen von drei unterschiedlichen Arthropoden-Hämocyaninen erstellt und
analysiert.
Für das 8x6-mere Hämocyanin des Pfeilschwanzes Limulus polyphemus wurden zwei
3D-Rekonstruktionen erstellt. Die erste 3D-Rekonstruktion besitzt eine Auflösung von
7,3 Å und stellt den deoxygenierten Zustand des respiratorischen Proteins dar. Die
zweite 3D-Rekonstruktion des 8x6-mers zeigt den oxygenierten Zustand des Proteins
und besitzt eine Auflösung von 7,9 Å. Die strukturellen Unterschiede zwischen beiden
3D-Rekonstruktionen wurden genauer untersucht und analysiert. Durch das flexible
Einpassen von Homologiemodellen der verschiedenen Untereinheitentypen in die
entsprechenden 3D-Rekonstruktionen konnten die Kontaktstellen, die zwischen den
Hexameren des 8x6-mers auftreten, genauer als früher beschrieben und erstmalig
Unterschiede beider Konformationszustände ermittelt werden. Für zwei Kontaktstellen
wurden molekulardynamische Simulationen durchgeführt, um Näheres über die
Interaktion der beteiligten Aminosäuren zu erfahren.
Für das 4x6-mer des Maulwurfskrebses Callianassa truncata wurde eine
3D-Rekonstruktion mit einer Auflösung von 4,9 Å erstellt. Diese erlaubte es, die
Kontaktstellen der vier Hexamere mit Hilfe von Homologiemodellen der beiden
Untereinheitentypen genauer zu untersuchen. Die vier hier beschriebenen
Kontaktstellen wurden zusätzlich mit Hilfe molekulardynamischer Simulationen
analysiert.
Die 3D-Rekonstruktion des 2x6-mers der Meeresassel Ligia oceanica erreichte eine
Auflösung von 8,6 Å und erbrachte neue Erkenntnisse über die ungewöhnliche
Assemblierung der beiden Hexamere und über die strukturelle Verwandtschaft der
verschiedenen Crustaceen-Hämocyanine. Arthropod hemocyanins are very large extracelluar respiratory proteins. They consist of several types of 75 kDa subunits, each containing a binuclear copper active site for reversible oxygen binding. The basic quaternary structure of arthropod hemocyanins is a hexamer (1x6-mer), but in the hemolymph of most species a characteristic type of oligo-hexamer predominates (2x6-mer, 4x6-mer, 6x6-mer, 8x6-mer). Many arthropod hemocyanins show a very high cooperativity of oxygen binding. In order to study this cooperativity at the level of single amino acids, and notably the allosteric force transfer at the inter-hexamer contacts, detailed information about the quaternery structure is required. 3D-electron microscopy of cryo-fixed proteins is the relevant method to determine the quaternary structure of such large proteins. In the present thesis I used this technique to reveal, in cooperation with others, the quaternary structure of three different arthropod hemocyanins in detail. In my first project, we established 3D-reconstructions of the oxy and the deoxy state of the 8x6-mer hemocyanin from the horseshoe crab Limulus polyphemus. The two 3D-reconstructions have resolutions of 7.3Å (deoxy state) and 7.9Å (oxy state), respectively. To analyze conformational changes at the inter-hexamer interfaces, atomistic homology models of the seven different subunit types were flexibly fitted into the corresponding 48 subunit positions of both 3D-reconstructions. The two resulting atomistic models of the 8x6-mer were then used to describe, for the first time at this structural level, conformational chances occuring during oxygenation and deoxygenation. Molecular dynamics simulations were applied to further substantiate the results. In my second project, we produced a 3D-reconstruction at 4.9Å resolution of the unique 4x6-mer hemocyanin from the ghost shrimp Callianassa truncata. This high resolution enabled producing, by flexible fitting, the first reliable atomistic homology model of this hemocyanin type. Additional molecular dynamics simulations revealed new insights in the structure and stability of the molecular interfaces between the hexamers of this 4x6-mer. Moreover, I obtained a 3D-reconstruction of the authentic 2x6-mer half-molecule which turned out to be very similar to the typical 2x6-mer of Brachyuran crabs. In my third project, we produced a 3D-reconstruction at 8,6Å resolution of the 2x6-mer hemocyanin from the marine isopod Ligia oceanica. Its unusual mode of inter-hexamer assembly, which is also found in Callianassa hemocyanin, is discussed in the framework of crustacean hemocyanin evolution. |
