Abstract: | Etablierung von Expressionsystemen für Gene der
Indolalkaloid-Biosynthese unter besonderer
Berücksichtigung von Cytochrom P450-Enzymen
In der vorliegenden Arbeit wurden Enzyme aus der Arzneipflanze Rauvolfia
serpentina bearbeitet. Es wurde versucht, das an der Biosynthese des Alkaloids
Ajmalin beteiligte Cytochrom P450-Enzym Vinorin-Hydroxylase heterolog und
funktionell zu exprimieren.
Ein zunächst unvollständiger, unbekannter Cytochrom P450-Klon konnte
komplettiert und eindeutig mittels heterologer Expression in sf9-Insektenzellen
als Cinnamoyl-Hydroxylase identifiziert
werden. Die Tauglichkeit des
Insektenzellsystems für die Untersuchung der Vinorin-Hydroxylase ist auf
Grund der deacetylierenden Wirkung der Insektenzellen auf das Substrat
Vinorin nicht gegeben.
Im Rahmen des Homology Cloning Projektes konnten mehrere Volllängenklone
und diverse Teilsequenzen von neuen Cytochrom P450-Klonen ermittelt
werden. Ausserdem wurde durch das unspezifische Binden eines degenerierten
Primers ein zusätzlicher Klon gefunden, der der Gruppe der löslichen
Reduktasen zugeordnet werden konnte. Diese putative Reduktase wurde auf
die Aktivität von mehreren Schlüsselenzymen der Ajmalin-Biosynthese durch
heterologe Expression in E.coli und anschliessende HPLC-gestützte
Aktivitätstests ohne Erfolg geprüft.
Bedingt durch die Untauglichkeit des Insektenzellsystems für die Identifizierung
der Vinorin-Hydroxylase, wurde ein neuartiges Modul-gestütztes, pflanzliches
Expressionsystem etabliert, um vorhandene P450-Volllängenklone auf Vinorin-
nHydroxylaseaktivität testen zu können. Die Funktionalität des Systems konnte
durch die heterologe Expression der Polyneuridinaldehyd Esterase bestätigt
werden. Trotzdem war es bis jetzt nicht möglich, die Cinnamoyl-Hydroxylase als
Kontrollenzym für das pflanzliche System oder aber die gesuchte Vinorin-
Hydroxylase in aktiver Form zu exprimieren. "Establishing of expressionsystems for genes of the indolalkaloide biosynthesis in consideration of P450s” The actual thesis is about the working with enzymes of the medicinal plant Rauvolfia serpentina. It was tried to investigate the vinorine-hydroxylase (VH) which is a P450 enzyme and catalizes the hydroxylation of the substrate vinorine. It was possible to complete a fragment of a unknown p450 clone and to identify as cinnamicacid-hydroxylase by heterologous expression in sf9-insect cells with the nbaculovirussystem. It is not possible to test P450 clones for VH-activity because of the deacetylation of the vinorine thru the insectcells. The result of the homology cloning were several parts of sequences of P450-clones and three fulllengthclones. With the unspecific binding of degenerated primers it was found the sequence of a unknown soluble reductase of R. serpentina. The identity of this reductase is still not known nevertheless the testing for the activity of some important reductases of R. serpentina. It was also possible to establish a new plant expression system which is based on the disposition of different modules to synthesis the final expression vector, the recombinant Tobacco Mosaic Virus, in the plant cell after infiltration with Agrobacterium tumefaciens. This expressionsystem was used to try the expression of the desirded P450 clones in Nicotiana benthamiana which is the host plant of the system. The functionality of the system was proved with the expression of the polyneuridine aldehyd esterase of Rauvolfia serpentina. Up to now it was not possible to express the cinnamic acid hydroxylase in a active way as a positive control enzyme for the expression of P450s. Also it was not possible to express the VH in a active way. |
