Title: Identifizierung eines adaptiven antibakteriellen Abwehrmechanismus des marinen Schwammes Suberites domuncula

Abstract: Schwämme (Porifera) sind die phylogenetisch ältesten Metazoa. Sie besitzen komplexe Abwehrsysteme, welche vor allem auf der Synthese bioaktiver niedermolekularer Sekundärmetaboliten beruhen und diese Tiere zu einer der reichhaltigsten Quellen für medizinisch nutzbare Wirkstoffe machen. Besonders der marine Einsiedler-Korkschwamm (Suberites domuncula) hat sich in den letzten Jahren zur Untersuchung der molekularen Zusammenhänge dieser Abwehrmechanismen als besonders geeignet herausgestellt. So wurden in diesem Schwamm beispielsweise zwei lyso-PAF (plättchenaktivierender Faktor) Derivate (1-O Hexadecyl-sn-glycero-3-phosphatidylcholin und 1-O-Octadecyl-sn-glycero-3-phosphatidylcholin) identifiziert und charakterisiert, sowie deren ausgeprägte antibakterielle Aktivität besonders gegenüber gramnegativen Bakterien demonstriert. Eine Behandlung mit der Modellsubstanz zur Simulation einer bakteriellen Infektion, dem Endotoxin Lipopolysaccharid (LPS), für insgesamt 72 Stunden resultierte in einem Anstieg der Expressionslevel zweier an der Biosynthese dieser bioaktiven Etherphospholipide beteiligten Proteine. Unter Anwendung der Methode des Differential Display konnte einerseits das Schlüsselenzym der Etherphospholipid Biosynthese Alkyl- Dihydroxyacetonphosphat (DHAP)-Synthase und andererseits die regulatorische Untereinheit der PAF-deacetylierenden PAF Acetylhydrolase I , dem Lissenzephalie 1 Protein, als adaptiv hochreguliert identifiziert werden. Beide Schwammproteine zeigten die charakteristischen Merkmale der Enzyme von Metazoa. Um den Zusammenhang zur adaptiven Synthese bioaktiver lyso-PAF Derivate herzustellen, wurde in der Folge eine Quantifizierung der Biosynthese Endprodukte durchgeführt. Dabei konnte mittels LC-MS Methodik ebenfalls ein adaptiver Konzentrationsanstieg für 1-O Hexadecyl-sn-glycero-3-phosphatidylcholin um 26,7 % und für 1-O-Octadecyl-sn-glycero-3-phosphatidylcholin um 25 % nach 72-stündigem LPS Kontakt nachgewiesen werden. Erstmals wurde somit sowohl auf der Basis der Analyse der differentiellen Genexpression zweier an der Etherphospholipid-Biosynthese beteiligten Proteine mittels der Methode des Differential Displays als auch auf der Basis der Quantifizierung der bioaktiven Biosynthese Endprodukte eine adaptive antibakterielle Reaktion des Schwammes Suberites domuncula nachgewiesen.
Sponges (Porifera) represent the evolutionary oldest metazoan phylum still extant today. They have developed a complex Bauplan, based on the existence of structural and regulatory molecules; many of these have been cloned and analyzed in the past years. The demosponge Suberites domuncula has been used as a suitable model to demonstrate that these animals not only possess an adaptive immune response on the level of cytokines, but also, as pointed out here, on the level of synthesis of bioactive alkyl-lipid derivatives. From specimens of S. domuncula the two lyso-PAF (platelet-activating factor) compounds, 1-O hexadecyl-sn-glycero-3-phosphocholine and 1-O-octadecyl-sn-glycero-3-phosphocholine, have been identified and characterized which show pronounced anti-bacterial activity. Treatment of the tissue with the model compound, the endotoxin lipopolysaccharide (LPS) for 72 h substantially increased the expression level of two key enzymes of the lyso PAF pathway, i.e. alkyl-dihydroxyacetonephosphate synthase (ADS) and the lissencephaly 1 gene. The latter one is a regulatory subunit of PAF acetylhydrolase I, which leads to lyso PAF formation. The results were obtained by application of the differential display technique. Both cDNAs where cloned from S. domuncula and the results were verified by northern blotting analysis. The sponge enzyme as well as the regulatory subunit comprise the characteristic features of metazoan enzymes; they are increasingly expressed in the tissue after exposure to LPS. In order to prove, that the increase of expression levels might result in an increasing concentration of the bioactive endproducts of etherlipid biosynthesis identified in S. domuncula, a quantification was performed. By application of an LC MS method, the increase of etherlipid concentrations due to endotoxin treatment was up to 26,7 % for 1-O-hexadecyl-sn-glycero-3-phosphocholine and up to 25 % for 1-O octydecyl-sn-glycero-3-phosphocholine and in comparison to non incubated sponge tissue. In conlusion, the increase of the synthesis levels of bioactive nonproteinaceous and low-molecular weight lipid derivatives was identified on the level of expression of proteins involved in their biosynthesis as well as on the level of quantification of the biosynthesis endproducts, namely the identified antibacterial lyso-PAF congeners.
