Abstract: | Untersuchungen zur posttranslationalen präS-Translokation des großen Hüllproteins des Hepatitis-B-Virus.
Das große (L) Hüllprotin des Hepatitis-B-Virus (HBV) besitzt die ungewöhnliche Eigenschaft,
mittels partieller, posttranslationaler Translokation seiner präS-Domäne durch intrazelluläre Membranen zwei unterschiedliche Transmembrantopologieen auszubilden. Unter Berücksichtigung der Hypothese eines HBV-spezifischen Transmembrankanals, der sich möglicherweise während der Virusmorphogenese bilden und die präS-Translokation ermöglichen könnte, wurden Parameter untersucht, welche die L-Topologie beeinflussen. Dazu wurden Wildtyp-L-Proteine und L-Mutanten in Säugerzellen synthetisiert und deren Topologie mittels Proteaseschutzversuchen untersucht. Ich konnte zeigen, daß alle Faktoren, für die angenommen wurde, daß sie für die Ausbildung einer HBV-spezifischen Pore und die damit verbundene präS-Reorientierung wichtig seien, entbehrlich sind. Im einzelnen konnte nachgewiesen werden, daß die posttranslationale präS-Translokation weder die Helferfunktion der HBV S und M Proteine, noch die kovalente Dimerausbildung der Hüllproteine benötigt. Weiterhin ergaben die Untersuchungen, daß keine der amphipathischen
Transmembrandomänen des L-Proteins an der präS-Reorientierung beteiligt ist. Vielmehr wurde die hydrophobe Transmembrandomäne 2 (TM2) als ausreichend und essentiell für diesen Prozeß identifiziert. Zellfraktionierungsstudien ergaben weiterhin, daß die präS-Reorientierung und damit die duale Topologie des L-Proteins innerhalb des Endoplasmatischen Retikulums (ER) herbeigeführt wird. Letztlich konnte eine Interaktion des L-Proteins mit zellulären Chaperonen (Hsc70, Hsp40, BiP) gezeigt werden, was eine Beteiligung dieser Proteine am Translokationsprozeß nahelegt. Analysis of the posttranslational preS-translocation of the hepatitis B virus large envelope protein. The large L envelope protein of the hepatitis B virus (HBV) has the peculiar capacity to form two transmembrane topologies via a yet uncharacterized process of partial posttranslational translocation of its preS domain across membranes. In view of a current model that predicts an HBV-specific channel generated during virion envelope assembly to enable preS translocation, I have examined parameters influencing L topogenesis by using protease protection analysis of wild-type and mutant L proteins, synthesized in transfected cells. I could demonstrate that, all determinants, thought to be responsible for channel formation, are dispensable for preS reorientation. In particular, I observed that this process does not require (i) the helper function of the HBV S and M envelope proteins, (ii) covalent dimer formation of envelope chains, and (iii) either of the three amphipathic transmembrane (TM) segments of L. Rather, the most hydrophobic TM2 of L segment was identified as a vital topogenic determinant, essential and sufficient for posttranslational preS translocation. Cell fractionation studies revealed that preS refolding and thus the dual topology of L is established at the endoplasmic reticulum (ER) membrane, and coimmunoprecipitation studies showed that the L protein interacts with cellular chaperones, like Hsc70 and BiP. |
