Title: Synthesen von Liganden zur Entwicklung eines in vitro Assays zur Untersuchung der Bindung an Humanem Serum-Albumin

Abstract: Titrations- und Kompetitionsexperimente, Gleichgewichtsdialysen und Job-Plots zeigten, dass eine fluoreszierende Fettsäure (NBD-FA) als spezifischer Marker für eine hochaffine Fettsäurebindestelle an HSA geeignet ist. Radioaktive Gleichgewichtsdialysen zeigten, dass der Fluorophor die Bindungsaffinität reduziert, die spezifische Bindung der Fettsäure aber nicht verhindert. Ein positiver Fluoreszenz-Resonanzenergietransfers lieferte den ersten Hinweis auf die Bindestelle von NBD-FA an HSA (Sudlow-Site I). Durch die Cokristallstruktur von NBD-FA mit HSA wurde Sudlow-Site I als Bindestelle identifiziert. Sulfasalazin ist kompetitiv zu NBD-FA. Zur Untersuchung der neu identifizierten Bindestelle wurden verschiedene Sulfasalazin-Grundkörper sowie N-Alkyl und N-Aryl substituierte Sulfasalazin-Derivate synthetisiert. Erste Struktur-Affinitäts-Beziehungen wurden durch Kompetitionsexperimente mit NBD-FA erhalten. Nach Identifizierung der Bindestelle von NBD-FA wurde ein trichromatischer Assay entwickelt. Als spezifische Sudlow-Site I und II-Marker wurden ein 4-Hydroxywarfarin und ein Dimethylamino-BODIPY entwickelt. Da die Ergebnisse aus den monochromatischen Assays mit dem neu entwickelten trichromatischen Assay reproduziert werden konnten, ermöglicht dieser Assay die simultane Untersuchung von drei Bindestellen an HSA. Die genaue Affinität für eine spezifische Bindestelle kann mit Hilfe des monochromatischen Assays bestimmt werden. Weiterhin wurden zur Entwicklung eines Sensors für glykierte HSA Versuche zur Synthese von Boronsäure-funktionalisierten Sulfasalazin-Derivaten unternommen. Dabei wurde eine dimere Struktur erhalten. Die Ringöffnung scheint mit einer Glucose-, Fructose- oder Glycerol-Lösung möglich zu sein.
A fluorescent fatty acid (NBD-FA) is a suitable sensor for a high affinity fatty acid binding site on HSA which was shown by titration und competition experiments, equilibrium dialysis and Job Plots. The binding affinity of the fatty acid decreases, but the fluorophore does not prevent specific binding. Sudlow Site I was identified as binding site by crystallography. Sulfasalazin is competitive to NBD-FA. To investigate the new identified binding site different sulfasalazin derivatives were synthesized. First structure-affinity relationships were established. The identification of the binding site of NBD-FA enabled the development of a trichromatic competition assay using 4-hydroxywarfarin and dimethylamino-BODIPY as site-specific sensors. The results of the monochromatic assays were reproduced. Thus a simultaneous determination of three independent binding sites on HSA can be conducted. Furthermore, attempts for the synthesis of boronic acid functionalized sulfasalazin derivatives for the development of a sensor for glycated HSA were performed. A dimeric structure was obtained which can be cleaved by glucose, fructose or glycerol solutions.
