Title: Die Interaktion zwischen OMI/HtrA2 und NG2 in oligodendroglialen Vorläuferzellen : NG2 und Homöostase?

Abstract: Myelinisierende Oligodendrozyten im ZNS entwickeln sich aus NG2-positiven OPC. NG2 ist ein Transmembranprotein der Zytoplasmamembran. Der intrazelluläre Teil besitzt ein PDZ-Bindemotiv am C-Terminus, das im Hefemodell (Y2H-Analyse) mit der PDZ-Domäne der mitochondrialen Serinprotease OMI/HtrA2 interagiert. OMI/HtrA2 ist unter normalen physiologischen Bedingungen in dem Intermembranraum der Mitochondrien lokalisiert. Unter oxidativem Stress bzw. während der Apoptose findet eine Translokalisation durch die permeabilisierte mitochondriale Membran in das Zytosol statt. Dort übt OMI/HtrA2 eine proapoptotische Funktion aus. Im Rahmen dieser Arbeit sollte die Interaktion von OMI/HtrA2 und NG2 in OPC unter physiologischen Bedingungen untersucht und die zelluläre Funktion geklärt werden. Die Ergebnisse der Y2H-Analyse konnten mit Co-Immunpräzipitationen verifiziert werden. Da die beiden Proteine unter normalen physiologischen Bedingungen räumlich getrennt sind, wurde in den OPC oxidativer Stress simuliert. Dadurch konnte die mitochondriale Membran permeabilisiert werden, was eine Voraussetzung für die Interaktion der beiden Proteine darstellt. Mit einem neu etablierten Zell-ELISA konnte gezeigt werden, dass unter oxidativen Stressbedingungen OMI/HtrA2 in das Zytosol translokalisiert wird und an NG2 bindet. Versuche mit OPC zeigten, dass NG2 eine antiapoptotische Funktion unter oxidativem Stress ausübt. Die Deletion des PDZ-Bindemotivs von NG2, über das OMI/HtrA2 gebunden wird, führt zu einer erhöhten Sensibilität gegenüber oxidativem Stress. Außerdem konnte mit einem spezifischen OMI/HtrA2-Inhibitor die erhöhte Anfälligkeit der NG2-defizienten Zellen annähernd ausgeglichen werden. Weiterhin wird die Proteaseaktivität von OMI/HtrA2 durch die Interaktion vermindert. Dadurch wird die proapoptotische Funktion von OMI/HtrA2 im Zytosol gehemmt. Zusammengefasst reduziert die Interaktion von OMI/HtrA2 und NG2 die Apoptoserate in OPC unter oxidativem Stress. In der NG2-Knockout Maus konnte eine gesteigerte Gliose-Entwicklung mit zunehmendem Alter beobachtet werden, die möglicherweise mit dem Verlust der protektiven Funktion von NG2 in Verbindung steht.
Myelinating oligodendrocytes in the CNS develop from NG2-positive OPC. NG2 is a transmembrane protein of the cytoplasmic membrane. The intracellular part has a PDZ binding motif at the C-terminus, which interacts with the PDZ domain of the mitochondrial serine protease OMI/HtrA2 in the yeast model (Y2H analysis). OMI/HtrA2 is localised under normal physiological conditions in the intermembrane space of the mitochondria. Under oxidative stress or during apoptosis OMI/HtrA2 is translocated through the permeabilised mitochondrial membrane into the cytosol. There OMI/HtrA2 exerts a proapoptotic function. In this work the interaction of OMI/HtrA2 and NG2 in OPC under physiological conditions was investigated and the cellular function clarified. The results of the Y2H analysis could be verified with co-immunoprecipitations. The mitochondrial membrane can be permeabilised by oxidative stress, which is a prerequisite for the interaction of the two proteins. Using a modified cell ELISA Assay, OMI/HtrA2 was translocated into the cytosol under oxidative stress conditions and binds to NG2. Experiments with OPC showed that NG2 exerts an anti-apoptotic function under oxidative stress. The deletion of the PDZ binding motif of NG2, which binds OMI/HtrA2, leads to increased sensitivity to oxidative stress. In addition, the increased susceptibility of NG2-deficient cells could be approximately compensated with a specific OMI/HtrA2 inhibitor. Furthermore, the protease activity of OMI/HtrA2 is reduced by the interaction. This inhibits the pro-apoptotic function of OMI/HtrA2 in the cytosol. In summary, the interaction of OMI/HtrA2 and NG2 reduces the rate of apoptosis in OPC under oxidative stress. In the NG2 knockout mouse, increased gliosis development with age could be observed, possibly related to the loss of protective function of NG2.
