Title: Wichtige Faktoren für die Expression von Interleukin 9 in murinen Mastzellen

Abstract: Interleukin 9 ist ein pleiotropes Cytokin mit verschiedenen Funktionen innerhalb des Immunsystems. Zu den IL-9-abhängigen Erkrankungen zählen u. a. allergisches Asthma, entzündliche Darmerkrankungen und mikrobielle Infektionen. Neben Mastzellen, bilden auch T-Helferzellen, wie Th9-Zellen, das Cytokin IL-9. Auf zellulärer Ebene fungiert IL-9 als Wachstumsfaktor für Mastzellen und kann in aktivierten Mastzellen die Produktion verschiedener Cytokine induzieren. Vergleichende Untersuchungen über die Aktivierung von Mastzellen verschiedener Mausstämme durch proinflammatorische Signale, wie das Lipopolysaccharid R-LPS, zeigen eine erhöhte IL-9-Expression in BMMC aus BALB/c, verglichen mit Mäusen des genetischen C57BL/6-Hintergrundes. Ursache für die gesteigerte Expression des Cytokins IL-9 ist eine verstärkte Expression des NF-κB-Proteins c-Rel, das als Transkriptionsfaktor die IL 9 Produktion direkt verstärkt. Die höhere c-Rel-Aktivität bedingt außerdem eine Steigerung der Expression der Transkriptionsfaktoren IRF-4 und IRF-1 und des proinflammatorischen Cytokins IL-1β, die wiederum positiv auf die Transkription von IL-9 einwirken. Somit liegt in BMMC aus BALB/c-Mäusen ein Netzwerk aus Signalwegen vor, die in Abhängigkeit von der Aktivität des NF-κB-Faktors c-Rel eine positive Verstärkung der Expression des Cytokins IL-9 bewirken. Verschiedene Vorarbeiten an murinen Mastzellen konnten den Transkriptionsfaktor NFATc2 bereits als wichtigen Faktor für die Produktion von IL-9 identifizieren. Durch den Einsatz eines Mausmodells mit einer Mastzell-spezifischen NFATc1-Defizienz sollte die Funktion des verwandten Faktors NFATc1 in Mastzellen analysiert werden. Überraschenderweise wurde jedoch festgestellt, dass es bedingt durch einen kompensatorischen Effekt von NFATc2, nach Stimulation mit dem Kofaktor R-LPS zu erhöhten IL-9-Werten in den IgE kreuzvernetzten Mastzellen kommt. Von der gesteigerten NFATc2-Wirkung sind auch die Transkriptionsfaktoren IRF-4 und IRF-1 betroffen, sowie das proinflammatorische Cytokin IL-1β. Die erhaltenen Resultate zeigen, dass das gewählte Mausmodell zur Untersuchung der Funktion des NFAT-Faktors NFATc1 in Mastzellen nicht geeignet ist, da es zu einer kompensatorischen Übernahme der Aufgaben von NFATc1 durch den Transkriptionsfaktor NFATc2 kommt.
Interleukin 9 is a cytokine with pleiotropic functions within the immune system. In addition to its protective role with regard to tumor immunology and immunity against intestinal parasites, such as nematodes, IL-9 has pro-inflammatory functions in various diseases. IL-9-driven diseases include allergic asthma, inflammatory bowel disease, and infections with Respiratory Syncytial Virus. Besides mast cells, Th9 cells are also a source of the cytokine IL-9. At the cellular level, IL-9 serves as a growth factor for mast cells and enhances the production of diverse cytokines in activated mast cells. In the present work, important signals acting on the production of IL-9 in murine bone marrow-derived mast cells (BMMC) were investigated. BMMC derived from different mouse inbred strains were activated by IgE in the presence of proinflammatory signals lipopolysaccharide R-LPS or IL-1β. Upon activation, mast cells derived from BALB/c mice exhibit increased IL-9 expression, compared to the genetic C57BL/6 background. The present study shows an enhanced expression of NF-κB protein c-Rel, a transcription factor, which directly amplifies the production of IL-9. Additionally, c-Rel also provokes an increased expression of the transcription factors IRF-4 and IRF-1 and the proinflammatory cytokine IL 1β. All these factors boost the transcription of IL-9. Thus, in BMMC derived from BALB/c mice a network of signaling pathways exist, which cause, depending on the activity of the NF-kB factor c Rel, a positive reinforcement of the expression of IL-9. Previous studies on murine mast cells were able to identify transcription factors of the NFAT family as important factors for the production of IL-9. In order to investigate the role of NFATc1, a selective gene targeting approach was inevitable, as general NFATc1 deficiency causes demise in utero. By using a mouse model with mast cell-specific NFATc1 deficiency, the function of the factor NFATc1 in mast cells was analyzed. However, compensatory overexpression of NFATc2 in IgE crosslinked mast cells upon stimulation with the cofactor R-LPS resultes in elevated IL-9 values. Overexpression of NFATc2 in NFATc1-deficient BMMC also causes elevated levels of the transcription factors IRF-4 and IRF-1, and the proinflammatory cytokine IL-1β. Our results show that the selected mouse model is not suitable to investigate the function of NFATc1 in mast cells, as overexpression of NFATc2 overcompensate the lack of NFATc1.
