Title: Charakterisierung des DxxxQ-Phosphatasemotivs des Sauerstoffsensors NreB in Staphylococcus carnosus

Abstract: In Bakterien regulieren Zweikomponentensysteme die Stoffwechselwege abhängig von den die Bakterien umgebenden Stimuli. Die Signaltransduktion erfolgt mittels Phosphoryltransfer von der den Reiz detektierenden Sensorhistidinkinase auf den Antwortregulator, welcher die spezifische Zellantwort auslöst. Dabei ist die Aktivierung des Antwortregulators durch Phosphorylierung und auch dessen Inaktivierung durch Dephosphorylierung von entscheidender Bedeutung. Das Gleichgewicht von Phosphorylierung und Dephosphorylierung gewährleistet die korrekte Adaptation des Stoffwechsels an die Stimuli. In HisKA_3 Sensorhistidinkinasen wurde ein DxxxQ-Phosphatasemotiv identifiziert, dessen konservierte Aminosäuren in der Regulation der Kinase- und Phosphataseaktivität der Sensoren eine entscheidende Rolle spielen. Das fakultativ anaerobe Bakterium Staphylococcus carnosus reguliert die anaerobe Nitratatmung über das NreBC-Zweikomponentensystem und den Nitratsensor NreA. Der Sauerstoffsensor NreB enthält ebenfalls ein DxxxQ-Motiv, dessen Funktion für NreB in der Regulation der anaeroben Nitratatmung in vivo und in vitro untersucht wurde. Die Funktion des konservierten Aspartatrests NreB(D160) konnte aufgrund von sich widersprechenden in vivo und in vitro Ergebnissen nicht eindeutig bestätigt werden. Varianten zeigten in in vivo Reportergenstudien eine durch Nitrat stimulierbare wildtypische Aktivität, während sie in vitro keine Autophosphorylierungsaktivität hatten. Der konservierte Glutaminrest NreB(Q164) spielt dagegen eine Rolle in der Dephosphorylierung des Antwortregulators. In Varianten ist die Dephosphorylierung von NreC sowohl in vivo wie in vitro gehemmt. Zusätzlich zur Phosphataseaktivität des Sensors NreB wird die Dephosphorylierung des Antwortregulators teilweise auch durch eine intrinsische Phosphataseaktivität von NreC katalysiert. Der Nitratsensor NreA hat keinen Einfluss auf die Dephosphorylierung von NreC. Der Phosphoryltransfer im NreBC-Zweikomponentensystem erfolgt von NreB(H159) auf NreC(D53). In vivo Reportergenstudien weisen auf eine zusätzliche, NreB(H159) unabhängige Aktivierung des Systems hin, während durch in vitro Phosphorylierungen des Antwortregulators NreC mit dem niedermolekularen Phosphodonor Carbamoylphosphat der konservierte Aspartatrest NreC(D53) als einziges Phosphorylierungsziel identifiziert wurde. Als Alternative zu Phosphorylierungsstudien mittels radioaktiv markiertem [γ-33P]-ATP wurde der Nachweis mittels phospho-affinem Phos-tag etabliert.
In bacteria two-component systems regulate metabolic pathways depending on different stimuli. The signal transduction takes place by means of phosphoryl transfer from the sensor histidine kinase to the response regulator, which triggers the specific cell response. The activation of the response regulator by phosphorylation and its inactivation by dephosphorylation is of crucial importance. The balance of phosphorylation and dephosphorylation ensures the correct adaptation of the metabolism to the stimuli. In HisKA_3 sensor histidine kinases, a DxxxQ phosphatase motif has been identified, whose conserved amino acids play a crucial role in the regulation of the kinase and phosphatase activity of the sensors. The facultative anaerobic bacterium Staphylococcus carnosus regulates anaerobic nitrate respiration via the NreBC two-component system and the nitrate sensor NreA. The oxygen sensor NreB contains a DxxxQ motif whose function for NreB has been studied in the regulation of anaerobic respiration in vivo and in vitro. The function of the conserved aspartate residue NreB(D160) could not be clearly confirmed due to contradictory results. In vivo, variants of this residue had a wild-typ phenotype whereas there was no detectable in vitro autophosphorylation activity. The conserved glutamine residue NreB(Q164) is involved in the dephosphorylation of the response regulator NreC. In variants the dephosphorylation of NreC is inhibited both in vivo and in vitro. In addition to the phosphatase activity of the NreB sensor, dephosphorylation of the response regulator is also catalyzed by an intrinsic NreC phosphatase activity. The nitrate sensor NreA has no influence on the dephosphorylation. Phosphoryl transfer in the NreBC two-component system occurs from NreB(H159) to NreC(D53). In vivo reporter gene studies indicate an additional NreB(H159) independent activation of the system, while in vitro phosphorylation of NreC with the low molecular weight phospho-donor carbamoyl phosphate identified the conserved aspartate residue NreC(D53) as the sole phosphorylation target. As an alternative to phosphorylation studies using radioactively labeled [γ-33P]-ATP, a detection system using Phos-tag was established.
