Zusammenfassung/Abstract: Der MAPK-Signalweg spielt eine zentrale Rolle in der Zellproliferation. Die Signaltransduktionskette reguliert Rezeptorsignale von der äußeren Zellmembran zum Zellkern, wo schließlich die Genexpression durch signalwegaktivierte Transkriptionsfaktoren reguliert wird. Mutationen in diesem Signalweg führen zu unkontrollierter Proliferation und sind in vielen Tumoren zu finden. Mutiertes Ras-Protein, welches die aktivierte Form, Ras-GTP, stabilisiert, führt zu kontinuierlicher Stimulation des Signalweges und damit zu unkontrolliertem Zellwachstum und ist in 30 % aller Tumoren zu finden. Um möglichst schnell potentielle, neue Medikamente zur Krebstherapie zu finden, ist es daher sinnvoll, diesen Signalweg in Zellen zu visualisieren. Deshalb wurde in dieser Arbeit ein fluoreszenzbasierter Zellassay entwickelt, welcher die Aktivität des Ras/Raf-Signalwegs direkt detektieren kann. Hierzu wurden Transfektionsvektoren erstellt, in denen die CRAF-cDNA mit dem Gen für EGFP bzw. mCherry gekoppelt wurde. Durch Transfektion von MDCK und MDCK-F3 Zellen mit den erstellten c-Raf-Fluorophor-Konstrukten konnten mehrere, stabil exprimierende Klone selektiert und kultiviert werden. Nach Charakterisierung dieser Zellklone, wurden die jeweils am besten geeigneten für fluoreszenzmikroskopische Aktivierungs- bzw. Inhibierungsversuche verwendet. Die Fusion eines Fluoreszenzproteins an das Ras-Effektorprotein c-Raf verhilft somit zur direkten mikroskopischen Detektion eines aktivierten Ras/Raf-Signalwegs in Zellen. Dieses Detektionssystem kann in Zukunft zur Wirkstofffindung beitragen. Auch der Wnt-Signalweg, welcher in vielen unterschiedlichen zellulären Prozessen wie Differenzierung, Proliferation und Apoptose involviert ist, kann bei Fehlregulation zur Tumorentstehung führen. Daher wurde die prinzipielle Möglichkeit der Herstellung einer sowohl Raf/Ras- als auch Wnt-Signalwegaktivität anzeigenden Doppeltransfektanten verifiziert. Des Weiteren wurde herausgefunden, dass Proliferationsergebnisse von BrdU und MTT-Assays in Wnt-3a-stimulierten HEK293T-Zellen wenig vergleichbar sind. Die Differenzen der beiden Proliferationsassays lassen sich durch die unterschiedlichen Anwendungspunkte erklären, nämlich einerseits die mitochondriale Aktivität (MTT) und andererseits die Synthese von DNA (BrdU). Um endgültige Beweise für die zelluläre Proliferationsrate zu bekommen, könnte die Interpretation der Ergebnisse einer einzigen Methode irreführend sein. In Anbetracht dessen ist es zu empfehlen stets mehrere Methoden zu verwenden.
The MAPK signaling pathway is a key cellular proliferation pathway that transmits a signal from a receptor on the cell surface to the DNA in the nucleus of the cell. Some components of this pathway are mutated or aberrantly expressed in many human cancers. Mutant Ras lacks GTPase activity and remains active leading to continuous stimulation of the signaling pathway and as a result to uncontrolled cell growth. To find new drugs for cancer therapy as quickly as possible, it is useful to visualize the MAPK pathway in cells. Therefore, a fluorescence-based cell assay was developed in this work, which can detect the activity of the MAPK pathway directly. For this purpose, transfection vectors were created in which CRAF cDNA was coupled to EGFP or mCherry. By transfecting MDCK and MDCK-F3 cells with the c-Raf fluorophore constructs, several, stably expressing clones could be selected and cultured. After characterization of these cell clones, the most suitable clones were used for fluorescence microscopic activation or inhibition experiments. In conclusion, by the use of fluorescent reporters fused to the Ras effector protein c-Raf the activity state of the MAPK signaling pathway can be visualized. This allows the analysis of the pathway and its protein-protein interactions as well as specific screenings for MAPK pathway modulating substances as potential drugs in the drug development for cancer treatment. The Wnt signaling pathway, which is involved in many essential cell processes, can also lead to tumorigenesis if dysregulated. For that reason, the basic possibility of producing dual transfectants indicating both MAPK and Wnt signaling pathway activity was verified. Furthermore, in this work was demonstrated that proliferation results of BrdU and MTT assays are hardly comparable in Wnt-3a stimulated HEK293T cells. The differences between the two proliferation assays can be explained by the different points of application, on the one hand the mitochondrial activity (MTT) and on the other hand the synthesis of DNA (BrdU). To get final evidence of the cellular proliferation rate, the interpretation of the results of a single method could be misleading. In consequence, it is highly recommended to use always several methods.
