BLaER1 macrophage model for the investigation of RNA stimulation and viral infections

| dc.description.abstract | During infections, the endolysosomal Toll-like receptor (TLR8) in macrophages of the innate immune response recognizes foreign RNA and triggers a signaling cascade leading to the release of cytokines and immune system activation. However, TLR8 stimulation is inhibited by 2’-O-methylation (2’-O-me), which is a known marker of eukaryotic RNA, through an unknown mechanism. To elucidate the molecular processes upstream of TLR8, we identified 2’-O-me-dependent RNA-binding proteins using the BLaER1 macrophage model and mass spectrometry-based proteomics. In this context, TRMT61A demonstrated increased binding to unmodified RNA, thereby suggesting a potential role in RNA processing prior to TLR8 activation. Moreover, studies of the subcellular localization of RNases and work by our collaborators confirmed the role of RNase6 in 2’-O-me-dependent processing of bacterial RNA as prerequisite for TLR8 activation. Furthermore, we characterized the BLaER1 macrophage model by analyzing proteomes during transdifferentiation and established it as model for orthoebolavirus infections. We demonstrated the susceptibility and permissiveness of BLaER1 to the virus and, using time-resolved proteome and secretome data, identified host factors that could explain orthoebolavirus pathogenicity and modulation of the immune response. Overall, our results establish BLaER1 macrophages as versatile model to studying RNA recognition and to expand our understanding of orthoebolavirus pathogenicity. | en_US |
| dc.description.abstract | Als Teil der angeborenen Immunantwort erkennt der endolysosomale Toll-like-Rezeptor 8 (TLR8) in Makrophagen während Infektionen fremde RNA und löst eine Signalkaskade aus, welche die Ausschüttung von Zytokinen und Aktivierung des Immunsystems verursacht. Allerdings wird die Aktivierung von TLR8 durch die 2‘-O-Methylierung (2‘-O-me), einen Marker von eukaryotischer RNA, durch einen unbekannten Mechanismus gehemmt. Zu Aufklärung der molekularen Vorgänge stromaufwärts von TLR8 identifizierten wir mithilfe des BLaER1-Makrophagenmodells und Massenspektrometrie-basierter Proteomik 2‘-O-me-abhängige RNA-Bindeproteine. Dabei zeigte TRMT61A eine erhöhte Affinität für unmodifizierte RNA und spielt damit einen potenzielle Rolle bei der RNA-Prozessierung vor TLR8-Aktivierung. Weiterhin bestätigten Untersuchungen der subzellulären Lokalisierung von RNasen und Arbeiten unserer Kollaborationspartner die Rolle von RNase6 bei de 2‘-O-me-abhängigen Prozessierung bakterieller RNA als Voraussetzung für die TLR8-Aktivierung. Darüber hinaus charakterisierten wir das BLaER1-Makrophagen-Modell mit der Analyse von Proteomen während der Transdifferenzierung und etablierten es als Modell für Orthoebolavirus-Infektionen. Dazu wiesen wir die Empfänglichkeit und Permissivität von BLaER1 für EBOV nach und identifizierten durch zeitaufgelöste Proteom- und Sekretomdaten verschiedene Wirtsfaktoren, welche die Pathogenität von Orthoebolaviren und die Modulation der Immunantwort durch das Virus erklären können. Insgesamt etablieren unsere Ergebnisse BLaER1-Makrophagen als vielseitiges Modellsystem zur Untersuchung der RNA-Erkennung im angeborenen Immunsystem und erweitern das Verständnis der Pathogenität von Orthoebolaviren. | de |
