dc.description.abstract | Das Nierenzellkarzinom gehört zu den zehn am häufigsten diagnostizierten Tumorarten in Deutschlan. Bei Diagnosestellung weisen bis zu 30 % der Patienten Metastasen auf und 40 % der Patienten mit initial lokal begrenzten Tumoren entwickeln im Verlauf der Erkrankung ein Rezidiv. Die systemische Therapie metastasierender Nierenzellkarzinome beinhaltet zielgerichtete Therapeutika wie den anti-VEGF-Inhibitor Bevacizumab, den Tyrosinkinase-Inhibitor Sunitinib und den mTOR-Inhibitor Temsirolimus. Doch trotz dieser Möglichkeiten bleibt die Prognose in vielen dieser Fälle eingeschränkt. Auf der Suche nach neuen Therapieoptionen sind in jüngster Zeit Cyclin-abhängige Kinasen in den Fokus gerückt. Verantwortlich für viele zelluläre Prozesse, wie u. a. die Zellzykluskontrolle, spielen sie bei der Onkogenese vieler Tumorarten durch pathologische Veränderung eine entscheidende Rolle. Der Erfolg des CDK4/CDK6-Kinasen-Inhibitors Palbociclib in der Therapie des metastasierten Brustkrebses ist ermutigend und könnte beispielhaft für neuartige Medikamente auch beim metastasierten Nierenkrebs sein. Ähnlich wie Cyclin-abhängigen Kinasen sind auch die Proteine der E2F-Familie in vielen Tumoren durch Mutationen verändert und ursächlich an der Tumorentstehung beteiligt. Im Speziellen das ubiquitär vorkommende E2F1 ist in diversen Tumoren überexprimiert und fördert den Tumorprogress. Als wichtiger Regulator des Zellzyklus und aufgrund seiner Beteiligung am DNA-Reparatursystem der Zelle kommt ihm entscheidende Bedeutung bei der Zellproliferation zu. Die Bedeutung von CDK11 und E2F1 beim metastasierten Nierenkrebs ist bisher wenig erforscht und soll daher genauer untersucht werden. Das Ziel der vorliegenden Arbeit besteht darin, zunächst jeweils mittels siRNA-Transfektion die Expression von CDK11 und E2F1 zu supprimieren. Anschließend werden funktionelle Analysen zur Bestimmung von Proliferation, Apoptose-Sensitivität und Sensitivität gegenüber verschiedenen Therapeutika durchgeführt, sowie unterschiedliche Signalwege untersucht. | de_DE |
dc.description.abstract | Renal cell carcinoma is one of the ten most frequently diagnosed types of tumors in Germany. When diagnosed, up to 30% of the patients show metastases and 40% of the patients with initially localized tumors develop a relapse in the course of the disease. The systemic therapy of metastatic renal cell carcinomas includes targeted therapeutics such as the anti-VEGF inhibitor bevacizumab, the tyrosine kinase inhibitor sunitinib, and the mTOR inhibitor temsirolimus. Despite these possibilities, the forecast remains limited in many of these cases. Cyclin-dependent kinases have recently come into focus in the search for new therapy options. Responsible for many cellular processes, such as u. a. cell cycle control, they play a crucial role in the oncogenesis of many types of tumors through pathological changes. The success of the CDK4 / CDK6 kinase inhibitor palbociclib in the therapy of metastatic breast cancer is encouraging and could be an example of new drugs for metastatic kidney cancer. Similar to cyclin-dependent kinases, the proteins of the E2F family are mutated in many tumors and are causally involved in the development of the tumor. The ubiquitous E2F1 in particular is overexpressed in various tumors and promotes tumor progression. As an important regulator of the cell cycle and due to its participation in the DNA repair system of the cell, it is of crucial importance in cell proliferation. The importance of CDK11 and E2F1 in metastatic kidney cancer has so far been poorly understood and should, therefore, be examined in more detail. The aim of the present work is to suppress the expression of CDK11 and E2F1 by siRNA transfection. Then functional analyzes for the determination of proliferation, apoptosis sensitivity, and sensitivity to different therapeutic agents are carried out, and different signaling pathways are examined. | en_GB |
