Title: Untersuchungen zur asymmetrischen Totalsynthese von Cripowellin

Abstract: Im Rahmen dieser Arbeit konnten wichtige Schritte auf dem Weg zur asymmetrischen Totalsynthese des Cripowellin-Aglycons gemacht und wichtige Erkenntnisse gewonnen werden. Es gelang dabei, einen Teil des Grundgerüsts des Kerns mit dem entscheidenden stereogenen Zentrum optisch aktiv aufzubauen. Die Aza-Claisen-Umlagerung diente hierbei als Schlüsselschritt zum stereoselektiven Aufbau eines Epoxids. Für die Aza-Claisen-Umlagerung dienten ein Allylamin und ein Säurefluorid als Ausgangsverbindungen. Das Allylamin konnte ausgehend von Piperonylalkohol über sechs Stufen (Schutzgruppen-operationen, Bromierung, metallorganische Reaktionen, Reduktion) mit einem Auxliar versehen und mit einer Gesamtausbeute von 56 % synthetisiert werden. Die Synthese des Säurefluorids konnte im Rahmen dieser Arbeit entscheidend verbessert werden, insbesondere bezüglich der Reinheit. Die Aza-Claisen-Umlagerung zu einem Pentensäurederivat selbst konnte mit einer sehr befriedigenden Ausbeute von 91 % durchgeführt werden. Dabei entstanden zwei Diastereomere im Verhältnis 92:8. Kristallisationsversuche schlugen leider fehl, sodass der Konfigurationsbeweis noch aussteht. Dieses Pentensäurederivat konnte zunächst durch einen Reduktionsschritt und saure Abspaltung der Schutzgruppe und des Auxiliars in ein Benzylchlorid überführt werden, welches dann durch Umsetzung mit Ammoniak zu einem Benzazepinon reagierte, ein weiteres wichtiges Schlüsselintermediat in dieser Synthese. Mit diesem Benzazepinon wurden viele Versuche zur Doppelbindungsfunk-tionalisierung unternommen, so z. B. Dihydroxylierungen und Epoxidierungen. Auch wurde das Amid zum Amin reduziert und mit Pentensäure erneut funktionalisiert. Mit dem so gewonnenen Dien wurden erste Versuche unternommen, über eine Grubbs-Metathese das bicyclische System des Cripowellins aufzubauen, die jedoch scheiterten.
This thesis takes important steps towards the asymmetric total synthesis of the cripowelline aglycone and presents important findings. I have successfully built a part of the basic skeleton of the core with the crucial stereogenic center in an optically active fahsion. The aza-Claisen rearrangement was used here as a key step for the stereoselective synthesis of an epoxide. For the aza-Claisen rearrangement an allylamine and an acid fluoride were used as starting compounds. Starting with piperonyl alcohol, in a six-step process (protective group operations, bromination, organometallic reactions, reduction), the allylamine was endued with an auxiliary and synthesized with an overall yield of 56%. The synthesis of the acid fluoride was also improved significantly in the context of my work, particularly with regard to its purity. The aza-Claisen rearrangement into pentenoic acid derivative itself was carried out with a very satisfactory yield of 91%. Two diastereomers resulted, with a ratio of 92:8. Attempts of crystallization failed, unfortunately, so that proof of configuration is still pending. This pentenoic acid derivative could be transferred to a benzyl chloride by a reduction, cleavage of the protecting group and cleavage of the auxiliary. This benzyl chloride reacted with ammonia to a benzazepinone, another important key intermediate in this synthesis. With this benzazepinone a number of attempts were made to functionalize the double bond, such as dihydroxylations and epoxidations. In addition, the amide was reduced to the amine and functionalized again with pentenoic acid. With the diene thereby obtained first attempts were made to build the bicyclic system of the cripowelline via a Grubbs metathesis, but these failed.
