Abstract: | Die Behandlung des Nierenzellkarzinoms (NZK) ist aufgrund entstehender Resistenzen limitiert. Ein Erstlinientherapeutikum ist Sunitinib. Artesunat (ART) und Shikonin (SHI) aus der Traditionellen Chinesischen Medizin könnten vielversprechende Additiva darstellen. Sie zeigten bereits antitumorale Effekte. Daten zum NZK sind jedoch rar. In der vorliegenden Arbeit wurde daher die antitumorale Aktivität von ART und SHI in therapiesensitiven (parentalen) und Sunitinib-resistenten NZK-Zellen untersucht.
Parentale und Sunitinib-resistente Caki-1, 786-O, KTCTL-26 und A-498 Zellen wurden mit ART und SHI behandelt. Anschließend wurden das Zellwachstum, die Proliferation, das klonogene Wachstum, der Zellzyklus, verschiedene Zelltode (Apoptose, Ferroptose, Nekroptose, Nekrose) sowie die zugrundeliegenden molekularen Modulationen untersucht. Weiterhin wurden die Adhäsion, Migration und Chemotaxis sowie die Expression von Adhäsionsrezeptoren analysiert.
ART und SHI induzierten eine signifikante Reduktion des Tumorzellwachstums und der Proliferation in parentalen und Sunitinib-resistenten NZK-Zellen. Dies ging nach ART-Behandlung mit einem G0/G1-Phase-Arrest und zelltypspezifisch einer Ferroptose-Induktion einher. Unter SHI-Gabe waren primär ein G2/M-Phase-Arrest und eine Nekroptose-Induktion zu beobachten. Entsprechende molekulare Modulationen waren detektierbar. Beide Stoffe inhibierten zudem signifikant das klonogene Wachstum. ART hemmte weiterhin die Adhäsion, Migration und Chemotaxis der NZK-Zellen.
Die ART- und SHI-Behandlung induzierte sowohl in parentalen als auch therapieresistenten NZK-Zellen antitumorale Effekte. Beide könnten somit eine integrative Therapie für Patienten mit fortgeschrittenem NZK darstellen. Treatment of renal cell carcinoma (RCC) is limited due to emerging resistance. A first-line therapeutic agent is sunitinib. Artesunate (ART) and shikonin (SHI) from traditional Chinese medicine could be promising treatment additives. They already showed antitumor effects. However, data on RCC are rare. Therefore, in the present study, the antitumor activity of ART and SHI was investigated in therapy-sensitive (parental) and sunitinib-resistant RCC cells. Parental and sunitinib-resistant Caki-1, 786-O, KTCTL-26, and A-498 cells were treated with ART and SHI. Subsequently, cell growth, proliferation, clonogenic growth, cell cycle, various cell deaths (apoptosis, ferroptosis, necroptosis, necrosis) and underlying molecular modulations were investigated. Furthermore, adhesion, migration, chemotaxis and adhesion receptor expression were analyzed. ART and SHI significantly reduced tumor cell growth and proliferation in parental and sunitinib-resistant RCC cells. This was accompanied by G0/G1 phase arrest and cell type-specific ferroptosis induction after ART treatment. SHI administration was primarily associated with G2/M phase arrest and necroptosis induction. Corresponding molecular modulations were detectable. Both agents also significantly inhibited clonogenic growth. ART further inhibited adhesion, migration and chemotaxis of RCC cells. ART and SHI treatment induced antitumor effects in parental and therapy-resistant RCC cells. Thus, both could represent an integrative therapy for patients with advanced RCC. |
