dc.description.abstractAcetylcholine binding proteins (AChBP) occur in the hemolymph of molluscs and annelids. AChBP is structurally and functionally homologous to the extracellular ligand-binding domain of the nicotinic acetylcholine receptor (nAChR). The tropical freshwater snail Biomphalaria glabrata (Planorbidae) has a special AChBP, which forms 25 nm regular dodecahedron out of twelve pentamers. In the present study, a 3D-reconstruction with a resolution of 3.5 Å was generated by recombinant BgAChBP1-dodecahedrons dissolved in pure water solved by 3D-electron microscopy. Applying site directed mutagenesis and subsequent electron microscopy, the importance of certain amino acids for the contacts between the pentamers was investigated. Further evidence was gained on the existence of a dipentamer in BgAChBP2; This aspect could not be finally clarified. In addition, evidence has been gathered that recombinant BgAChBP1 binds amorphous calcium carbonate and is maybe involved in the formation of the snail shell. Recombinant BgAChBP2 appears to bind amorphous calcium carbonate with low affinity. In the native planorbid snail Planorbarius corneus AChBP dodecahedra were detected in the hemolymph and biochemically enriched. Transcriptome analysis provided three AChBP sequences, two of them were complete and the third was almost complete. As declined from phylogenetic and biochemical analysis, no protein in P. corneus has been found which is orthologous to BgAChBP1. The dodecahedra obviously has been formed by an AChBP similar to BgAChBP2. This suggests that the BgAChBP2 also forms dodecahedra in the haemolymph of B. glabrata. However the recombinantly expressed BgAChBP2 is (so far) unable to form dodecahedra. This could be due to a certain disulfide bridge, which could be crucial for the oligomerization of two pentamers in BgAChBP2.en_GB
dc.description.abstractAcetylcholin-Bindeprotein (AChBP) kommt in der Hämolymphe von Mollusken und Anneliden vor. AChBP ist strukturell und funktionell homolog zur extrazellulären ligandenbindenden Domäne des nikotinischen Acetylcholinrezeptors (nAChR). Die tropische Süßwasserschnecke Biomphalaria glabrata (Familie Planorbidae) besitzt als Besonderheit ein AChBP, das 25 nm große reguläre Dodekaeder aus zwölf Pentameren bildet. In der vorliegenden Arbeit wurden von rekombinanten, in reinem Wasser gelösten BgAChBP1-Dodekaedern vermittels 3D-Elektronenmikroskopie eine 3D-Rekonstruktion mit einer Auflösung von 3,5 Å erstellt. Vermittels gerichteter Mutagenese und anschließender Elektronenmikroskopie wurde die Bedeutung bestimmter Aminosäuren für die Kontakte zwischen den Pentameren untersucht. Zur Existenz eines Dipentamers beim BgAChBP2 wurden weitere Hinweise gesammelt; dieser Aspekt konnte jedoch nicht abschließend geklärt werden. Darüber hinaus wurden Hinweise gesammelt, dass rekombinantes BgAChBP1 amorphes Calciumcarbonat bindet und somit wahrscheinlich an der Bildung des Schneckenhauses beteiligt ist. Rekombinantes BgAChBP2 scheint amorphes Calciumcarbonat in weit geringerem Umfang zu binden. Bei der einheimischen planorbiden Schnecke Planorbarius corneus wurden AChBP-Dodekaeder in der Hämolymphe nachgewiesen und biochemisch angereichert. Eine Transkriptomanalyse lieferte drei AChBP-Sequenzen, von denen zwei vollständig und die dritte fast vollständig waren. Wie der molekulare Stammbaum und biochemische Analysen zeigten, wurde in P. corneus kein zum BgAChBP1 orthologes Protein gefunden. Die Dodekaeder gehen hier ganz offensichtlich auf ein AChBP zurück, das dem BgAChBP2 ähnlich ist. Das legt den Verdacht nahe, dass in der Hämolymphe von B. glabrata das BgAChBP2 ebenfalls Dodekaeder bildet und dass lediglich das rekombinant exprimierte BgAChBP2 dazu (bisher) nicht in der Lage ist. Dies könnte an einer bestimmten Disulfidbrücke liegen, die sich beim BgAChBP2 zwischen zwei Pentameren ausbilden müsste.de_DE
