Title: Synthese und Selbstorganisation von Perylendiimid-Oligonucleotid-Konjugaten

Abstract: Im Rahmen dieser Arbeit wurde eine Methode entwickelt, Perylendiimidfarbstoffe mit Oligonucleotiden in der Lösung zu verknüpfen. Das Ziel der Arbeit war die nicht-kovalente Synthese von Perylendiimid-DNA- und Protein- supramolekularen Strukturen. Dabei werden die molekularen Erkennungseigenschaften von DNA und Proteinen zunutze gemacht. Insgesamt drei Themenbereiche wurden dabei betrachtet: 1. Synthese und Hybridisierung von symmetrischen und asymmetrischen Perylendiimid-bis(oligonucleotid)-konjugaten für die Bildung supramolekularer Strukturen, 2. Erzeugung von Oberflächenstrukturen auf der Basis von Streptavidin-Perylendiimid-Komplexen, 3. Synthese wasserlöslicher Rylenfarbstoffe für Anwendungen in biologischen Systemen. Zur Synthese und Hybridisierung von Perylendiimid-Oligonucleotid-Konjugaten wurde eine neue Idee verfolgt und erfolgreich realisiert. Dabei handelt es sich um die Synthese von Perylendiimid-DNA-Polymeren durch nicht-kovalente Bindungen. Die Basis des entwickelten Konzepts ist die Ausnutzung der Erkennungseigenschaften der DNA, um Perylendiimidmoleküle in eine lineare Makrostruktur zu organisieren, was sonst nur durch komplizierte chemische Polymersynthese zugänglich wäre. Die Selbstorganisation von zwei komplementären Perylendiimid-bis(oligonucleotid)-konjugaten (PODN1 und PODN2), die an der 5`-Position verknüpft sind, führte zu einem linearen Perylendiimid-DNA-Polymer in der Form von …ABABABAB…., das mit Hilfe von Gelelektrophorese charakterisiert wurde. Eindrucksvoll war auch die erfolgreiche Kopplung des hydrophoben Perylendiimids mit zwei unterschiedlichen Oligonucleotidsequenzen in der Lösung, um asymmetrische Perylendiimid-bis(oligonucleotid)-konjugate zu synthetisieren. Mit solchen asymmetrischen Konjugaten konnte die programmierbare Selbstorganisation der Perylendiimid-Oligonucleotide zu einer definierten Polymerstruktur realisiert werden. Die Synthese von PDI-(biotin)2 wurde vorgestellt. Durch die spezifische Erkennungseigenschaft zwischen Biotin und Streptavidin ist es möglich, eine Oberflächenstruktur zu bilden. Die Immobilisierungsexperimente zeigten, dass das PDI (biotin)2 Streptavidin erkennen und binden kann. Dabei konnte eine multischichtige Nanostruktur (5 Doppelschichten) auf einer Goldoberfläche.
In this work a method has been developed for the solution-phase coupling of perylenediimide dyes to oligonucleotide. The aim of this thesis was the non-covalent synthesis of perylene-DNA- and protein-supramolecular structures. The molecular recognition properties of DNA and proteins were the basis to obtain such structures. Three topics were considered in this work: 1. Synthesis and Hybridization of symmetrical and asymmetrical Perylenediimide-bis(oligonucleotide)-conjugates for the construction of supramolecular structures 2. Manufacturing of surface architecture based on Streptavidin-Perylenediimide-complexes 3. Synthesis of water-soluble Rylene dyes for applications in Biological systems For the synthesis and hybridization of perylenediimide-oligonucleotid-conjugates a new idea has been worked out and realized. That was the construction of perylenediimide-DNA-polymers via non-covalent bonding. The basis of the developed concept was the utilization of the molecular recognition properties of DNA to organize perylenediimide molecules in a linear macrostructures, which could be only possible by using complicated chemical polymer synthesis. The Self-Assembly of two complementary perylenediimide-bis(oligonucleotide)-conjugates (PODN1 und PODN2), which are both linked at the 5`-position of the oligonucleotide, has led to a linear perylenediimide-DNA-polymer in the form of a block copolymer …ABABABAB. The polymer was characterized by polyacrylamide gel electrophoresis. Impressive was also the successful solution-phase coupling of the hydrophobic Perylenediimide with two different oligonucleotide sequences in order to synthesize asymmetric perylenediimide-bis(oligonucleotide)-conjugates. With the help of such asymmetric conjugates it was possible to self-assemble perylenediimide-oligonucleotides into a defined polymer structure. Also the synthesis of bisbiotinylated perylenediimide (PDI-(biotin)2) has led to the construction of a surface streptavidin-perylenediimide multi-layer structure, in which the molecular recognition properties of streptavidin and biotin have been utilized. The immobilization experiments have shown that PDI (biotin)2 has recognised and bound to streptavidin. A multi-layer nanostructure (5 double layers) on Ag/Au has been obtained.
