dc.description.abstractErhöhte Spiegel von oxidativem Stress bedingen Atherosklerose, eine Krankheit die über 50% aller Todesfälle in der westlichen Welt ausmacht. Es ist entscheidend Mechanismen zur Abwehr dieser Krankheit zu ergründen.rnDa genetische Polymorphismen des körpereigenen Enzyms Paraoxonase 2 (PON2) mit kardiovaskulären Erkrankungen assoziiert sind, wurden ihre Regulation und potentiell antioxidativen Funktionen in vaskulären Zellen analysiert. Mittels verschiedener molekularbiologischer Methoden konnte ich erstmals zeigen, dass PON2 in vaskulären Zellen vornehmlich subzellulär im ER lokalisiert ist. Anhand verschiedener Experimente wurde PON2 als potenter Faktor zur Reduktion von ROS identifiziert. Erhöhte ROS-Spiegel führen zur Aktivierung eines als unfolded protein response (UPR) bekannten ER-Stress-Signalwegs. Dieser ist neben Atherosklerose in eine Vielzahl von Erkrankungen involviert und hat kritischen Einfluss auf das Überleben oder Absterben von Zellen. Durchgeführte Promoter-Reporter Studien bewiesen die Induktion der Protein-Expression von PON2 nach Aktivierung des UPR-Signalwegs, was als kompensatorischer Mechanismus der Zelle zur Vermeidung UPR-induzierter Apoptose verstanden werden könnte. PON2 wehrt oxidativen Stress und die UPR-induzierte Apoptose ab und ist ein protektiver Faktor vor Atherosklerose.rnIn einem Krebsmodell könnte PON2 aber als antiapoptotischer Faktor entscheidend am Überleben von Tumorzellen beteiligt sein. Gerade diese beiden gegensätzlichen Aspekte der antiapoptotischen Funktion des Proteins zeigen die Notwendigkeit für weitere Untersuchungen zu PON2 auf.rnde_DE
dc.description.abstractElevated levels of reactive oxygen species (ROS) predispose to the development of atherosclerosis, a disease which is responsible for over 50% of morbidity and mortality in western societies. The fact that cardiovascular diseases cause a higher mortality than all cancers combined stresses the need to identify mechanisms to protect against this disease. rnBecause genetic polymorphisms of human paraoxonase-2 (PON2) have been associated with cardiovascular diseases, its regulation and potential antioxidative function were analyzed in this study. The analysis of the subcellular location by different techniques showed for the first time an association of PON2 mainly with the ER, mitochondria and nuclear membrane. Since enhanced levels of reactive oxygen species (ROS) are key features of atherosclerosis and other diseases, we determined PON2's effect on ROS and found that enhanced PON2 expression significantly reduced intracellular ROS levels. Elevated levels of oxidative stress lead to the activation of an ER-stress pathway, known as unfolded protein response (UPR), which is involved in many different diseases and has critical influence in survival or induction of apoptosis in cells. Our gene reporter experiments showed an increased PON2 promoter activity in response to UPR activation. This could be interpreted as a compensationary mechanism to avoid UPR-induced oxidative stress and / or apoptosis. Thus, PON2 defends vascular cells against oxidative stress and UPR-induced apoptosis and is a protective factor against atherosclerosis.rnFinally, given the importance of both ROS and ER stress in diseases other than atherosclerosis, in particular cancer, there is a possibility that the antiapoptotic qualities of PON2 play a major role for the survival of tumor cells. Future studies may therefore address these two antithetic aspects of PON2's antiapoptotic function, which may be appreciated from a cardiovascular standpoint, but detrimental when considering cancer development.rnen_GB
