Zusammenfassung/Abstract: Zusammenfassung:Die Quartärstruktur des respiratorischen Proteins Hämocyanin (Isoform HtH1) aus der marinen Schnecke Haliotis tuberculata wurde vermittels Kryoelektronen-mikroskopie und 3D-Rekonstruktion untersucht. Das Molekül ist zylinderförmig, hat einen Durchmesser von ca. 35 nm und besteht aus einer Zylinderwand und einem internen Kragenkomplex. Dieser wiederum besteht aus einem Collar und einem Arc.Die kryoelektronenmikroskopischen Aufnahmen von in glasartigem Eis fixierten HtH1-Molekülen brachte eine enorme Verbesserung der Anzahl der zur Verfügung stehenden Ansichtswinkel gegenüber den negativkontrastierten Molekülen, die auf Karbonfilm präpariert waren.Die 3D-Rekonstruktion des HtH1 mittels Aufnahmen bei drei verschiedenen Defo-kuswerten verbesserte die Auflösung noch einmal deutlich gegenüber den Rekon-struktionen, die aus Aufnahmen bei einem festen Defokuswert gemacht wurden, und zwar auf 12 Å. Das Molekül besitzt eine D5-Symmetrie.Aus dieser bisher genausten Rekonstruktion eines Molluskenhämocyanins aus EM-Bildern ließen sich folgende neue Strukturdetails ableiten:· Ein Untereinheitendimer konnte als Repeating Unit im Dekamer des HtH1 beschrieben werden.· Das Untereinheitendimer konnte aus der 3D-Dichtekarte isoliert werden. Es be-steht eindeutig aus 16 Massen, die funktionellen Domänen entsprechen. Zwei dieser Massen bilden den Collar, zwei den Arc und 12 das Wandsegment.· Die gegenläufige Anordnung der beiden Untereinheiten innerhalb dieses Unte-reinheitendimers konnten bestätigt und auf zwei Möglichkeiten eingeschränkt werden.· Die Zahl der alternativen Anordnungen der 16 funktionellen Domänen (HtH1-a bis HtH1-h) im Untereinheitendimer konnten von 80 auf 2 eingeengt werden.· Es konnte über molekulares Modellieren mithilfe einer publizierten Kristallstruk-tur eine 3D-Struktur fastatomarer Auflösung der funktionellen Domäne HtH1-g berechnet werden.· Die funktionelle Domäne HtH1-g konnte als Domänenpaar plausibel in die 3D?Dichtekarte des Untereinheitendimers eingepasst werden, und zwar in die beiden Massen des Arc.Aus der elektronenmikroskopisch gewonnenen Dichtekarte wurde mit Hilfe des
Abstract:The quaternary structure of the type 1 isoform of the extracellular respiratory hemocyanin (HtH1) from the marine gastropod mollusc Haliotis tuberculata has been examined by cryoelectron microscopy, with digital image analysis and 3-dimensional (3-D) reconstruction. The ca 800 MDa molecule, termed the HtH1 didecamer, is a hollow cylinder. It has an external diameter of ca 35 nm and a height of ca 36 nm, and consists of a thin fenestrated cylinder wall with an internal collar complex at each end, containing collar and arc components. Cryoelectron microscopic images from a large number of HtH1 molecules suspended in a thin layer of vitreous ice have provided an large improvement to the number of angular orientations available for image processing, compared to those previously obtained from negatively stained molecules spread on a carbon support film. The 3-D reconstruction from the initial HtH1 images, obtained at three different instrumental defocus levels with phase contrast transfer function (CTF) correction, which resulted by merging in an attainable 3-D resolution of 12 Å, a considerable improvement over what can be obtained from a single defocus level. The cylindrical HtH1 didecamer possesses D5-symmetry. From this improved reconstruction of a molluscan hemocyanin didecamer, obtained from cryoelectron micrographs, the following significant structure features have been shown: · The subunit dimer has been defined as the repeating unit in the decamer of the HtH1. · The subunit dimer has been isolated from within the 3-D reconstruction. It consists of two closely apposed sets of eight masses, which correspond to the eight immunologically-defined copper-containing functional units (FU's; abcdefgh) of each of the two subunits. Two of these FUs form the internal collar, two the arc and the remaining 12 FUs the cylinder wall. An anti-parallel arrangement of the two subunits is supported.· The anti-parallel arrangement of the two subunits within the subunit dimer now generates only two structural possiblites for the FU path within the dimer, to comply with the assymetrically located collar complex of the decamer.· Molecular modelling the 3-D structure of HtH1 FU-g , at close to atomic resolution, has been obtained by fitting the published crystal structure of Octopus dofleini FU-g as a reference.· The arc component of the HtH1 subunit dimer has been convincingly defined as a pair of FU's, by the fitting the FU-g from each of the two subunits into the 3-D density map.From the available electron microscopically determined 12 Å resolution density map, a solid model of the HtH1 didecamer has been generated by means of the industrial rapid prototyping procedure. This model possesses inherent regions of weakness that appear to represent the repeating zones of interaction between the more compact subunit dimers. Altogether, the data produced takes our understand of the quaternary structure of the molluscan hemocyanin a substantial step forward. However, it remains necessary to achieve a higher electron optical resolution for the a-helices and atomic detail within the FUs to be directly defined.
